Affinity-Based Assays

While proteomics data provide a global view of all proteins present in a sample, immunoassays can provide a more focused view on specific proteins of interest. PBL offers three immunoassay platforms that can be customized to each individual project: Olink PEA technology, Meso Scale Discovery and Quanterix™. All our assays are precise and highly sensitive, and our experts will help you choose the right one for you!

Olink Proximity Extension Assay (PEA) technology

The Olink PEA technology allows for the unbiased simultaneous quantification of multiple protein biomarkers in a single assay. It enables high-throughput quantification of proteins using minimal volumes (as low as 1µl) of serum, plasma, or any other type of biological sample. Less than a drop of blood is enough to measure the protein profile in a sample. The Olink Target PEA approach uses matched pairs of antibodies labeled with complementary DNA oligonucleotide tags to specifically bind to target proteins. When the antibodies bind to their target proteins, they bring the oligonucleotides within proximity to one another, allowing for hybridization and formation of a DNA “barcode” that is unique to each antibody pair. DNA barcodes are amplified via PCR to boost the signal-to-noise ratio of each target, and then quantified via qPCR to determine the target protein concentrations in samples.

Meso Scale Discovery (MSD) technology

The MSD technology allows for the absolute quantification of protein biomarkers by using multiplex, highly sensitive quantitative immunoassay platform. MSD assays are primarily sandwich assays, where the primary antibody targeting the protein of interest is immobilized on the surface of the microplate, while the detection antibody is conjugated with an electrochemiluminescent label for quantitative readout.

Electrochemiluminescence detection offers a unique combination of sensitivity and increased dynamic range, that cannot be matched by the classic ELISA assay. The MSD Multi-Spot microplates can efficiently multiplex up to 10 assays per sample, making MSD a high-throughput platform for multiplex biomarker quantification.

Quanterix™ Single Molecule Array™ (SIMOA™) technology

This Quanterix™ ‘digital ELISA’ bead technology allows for detection and quantification of low-abundance proteins in complex biological samples. SIMOA™ immunoassay uses a unique microfluidic-based platform that allows for the detection of individual proteins in a sample, rather than relying on the traditional bulk measurements. Like traditional ELISAs, the SIMOA assay is a sandwich immunoassay. However, in this case the bead-based detection technology allows for the detection of even a single molecule of antibody-bound protein. The Quanterix™ platform can deliver 1000-fold higher sensitivity than traditional ELISAs, yielding analyte detection in the femtomolar range and enabling quantification of proteins historically difficult to detect, like CNS-specific or serum/plasma-derived biomarkers.
